چکیده
A better understanding of tumor infiltrating leukocyte (TIL) differentiation is necessary to advance therapeutic strategies and inform melanoma patient outcomes. We have previously identified that the transcriptome CD8+ TILs can be placed on a spectrum early activation late memory states. Halting in associated with poor response immune checkpoint blockade (ICB), whereas progression towards resident states was productive ICB responses improved survival However, it remains unknown which, if any murine models currently being used model T cell demonstrate this trajectory. Identification such would allow us study signals driving differentiation. queried public dataset where OT-1 cells were activated vitro transferred into B16-OVA bearing mice presence or absence ICB. found treated which exhibited control had upregulated gene programs positive patients, as well persistent signatures memory. Control nonresponse activation, demonstrating faithfully captured trajectory we for human TILs. Future work will extend analysis include other congenic varied immunogenicity YUMM, YUMMER, examine cues shift states, hopes identifying rational combinations

Expansion and Characterization of Human Melanoma Tumor-Infiltrating Lymphocytes (TILs)
BACKGROUND Various immunotherapeutic strategies for cancer are aimed at augmenting the T cell response against tumor cells. Adoptive cell therapy (ACT), where T cells are manipulated ex vivo and subsequently re-infused in an autologous manner, has been performed using T cells from various sources. Some of the highest clinical response rates for metastatic melanoma have been reported in trials u...
متن کاملTrends in Adoptive Tumor Infiltrating Lymphocytes (TILs) Therapy
Adoptive T cell therapy based on tumor infiltrating lymphocytes (TILs) is the most effective treatment for malignant melanoma. The TILs therapy has demonstrated high overall response rates, resulting in tumor regression and prolonged survival in comparison to IL-2 and ipilimumab treatments. Expansion of TILs under exvivo conditions using patients’ own tumor tissue is a prerequisite for effectiv...

BACKGROUND The morphological evaluation of tumor-infiltrating lymphocytes (TILs) in breast cancer (BC) is gaining momentum as evidence strengthens for the clinical relevance of this immunological biomarker. Accumulating evidence suggests that the extent of lymphocytic infiltration in tumor tissue can be assessed as a major parameter by evaluation of hematoxylin and eosin (H&E)-stained tumor sec...

9 Abstract 10 11 Tumor infiltrating lymphocytes (TILs) have been recognized in various 12 cancers and may reflect a host immune response to malignant cells. TILs 13 are a heterogeneous population of various types of mononuclear cells, 14 including CD8 or CD4 + T cells and their subsets, B cells, myeloid 15 derived suppressor cells (MDSC), macrophages, and other cells. Immuno16 suppressive facto...
متن کاملCancer Immunology: Automated Enumeration of Tumor-Infiltrating Lymphocytes (TILs) in Melanoma
In many cancers, tumor-infiltrating lymphocytes (TILs) indicate levels of tumor immunogenicity and are a strong predictor of survival. An understanding of the phenotype and spatial distribution of TILs within tumor regions would be advantageous for characterizing host response. However, visual TIL assessment is prone to error and multimarker quantitation is difficult with standard methods. Here...
